     1. Instruments and reagents

    1.1 high performance liquid chromatography: Shimadzu lc-16 high performance liquid chromatography; equipped with spd-16 UV detector

    1.2 ultrasonic cleaning instrument: power 250W, frequency 40KHz

    1.3 analytical balance: sensitivity 0.00001g

    1.4 volumetric flask (25ml, 10ml)

    1.5 pipette (5ml)

    1.6 sennoside B reference substance (99% purity)

    1.7 sennoside a reference substance (purity 98%)

    1.8 acetonitrile (Chromatographic grade)

    1.9 phosphoric acid (analytical grade)

    1.10 sodium bicarbonate

    2. Experimental steps

    2.1. Solution preparation

    2.1.1 preparation of reference solution

    Take an appropriate amount of sennoside A and sennoside B, dry them under reduced pressure for 12 hours, put them into a brown measuring flask, add 0.1% sodium bicarbonate solution to make a mixed solution containing 0.05 mg sennoside A and 0.1 mg sennoside B per 1 ml, and shake well to obtain the reference solution.

    2.1.2 preparation of sample solution

    The sample is about 0.05 g, accurately weighed, put into a conical flask with a stopper, add an appropriate amount of 0.1% sodium bicarbonate solution, ultrasonic treatment for 15 minutes (30 ~ 40 ), cool, use 0.1% sodium bicarbonate solution to fix the volume, shake well, filter, and take the filtrate to obtain the sample solution.

    2.2. Determination

    2.2.1 chromatographic conditions

    Chromatographic column: wondasil c18-wr 5um 4.6x250mm

    Column temperature: 30

    Flow rate: 1.0 L / min;

    Wavelength: 340nm

    Injection volume: 20 μ L;

    Mobile phase:

    A: Acetonitrile

    B: 0.1% phosphoric acid aqueous solution

    Gradient condition:

    Time [min] %A %B

    0 15 85

    15 25 75

    25 30 70

    30 80 20

    2.2.2 testing

    Under the above chromatographic conditions, after the instrument is stable and the baseline is stable, extract 20 μ l of the control solution and the test solution respectively, inject them into the liquid chromatograph for determination.

    5. Content calculation

    The content was calculated by external standard method.

    Sennoside content (%) = sennoside B + sennoside a
